Elironrasib (RMC-6291) marks a significant advancement in drug development by enabling direct targeting of the active, GTP-bound form of KRAS G12C, a molecular target long regarded as inaccessible to therapeutic intervention. 


The resulting ternary assembly effectively blocks downstream oncogenic signaling and drives tumor regression across a range of preclinical cancer models carrying KRAS G12C mutations. Through a structure-based medicinal chemistry optimization program, elironrasib was identified as a highly potent, orally available, and selective covalent inhibitor of RAS(ON) G12C. Elironrasib is currently under evaluation in multiple Phase 1 clinical studies (NCT05462717, NCT06128551, NCT06162221), where early clinical findings suggest meaningful activity in patients who have developed resistance to first-generation KRAS G12C inhibitors that target the inactive RAS state.
